One Dose of Psilocybin Reshapes the Brain for a Month, Study Finds

A single dose of psilocybin, the psychedelic compound in magic mushrooms, produced measurable changes in the human brain that were still detectable a full month after the experience, according to a study that offers new clues about why the drug shows promise for treating depression and other stubborn mental-health conditions.
Researchers at the University of California, San Francisco, and Imperial College London recruited 28 healthy volunteers who had never taken a psychedelic before. In a carefully controlled design, each participant first received a 1-milligram dose that served as a placebo, and then, a month later, a much larger 25-milligram dose intended to induce a full psychedelic experience. The team tracked brain activity throughout using EEG, which records electrical signals from the scalp.
The changes appeared quickly. Within 60 minutes of taking the high dose, participants' EEG readings showed a sharp spike in what scientists call brain entropy — a measure of how varied, flexible and unpredictable the brain's activity is from moment to moment. Rather than settling into its usual well-worn patterns, the brain under psilocybin explored a much wider range of states, an effect that researchers increasingly believe is central to how psychedelics work.
Crucially, the effects did not vanish when the trip ended. The study, published in the journal Nature Communications, found measurable changes in brain activity, and possible changes in brain structure, that persisted for up to a month after the single session. That durability is what makes psilocybin so intriguing to clinicians: unlike conventional antidepressants that must be taken daily, a single guided dose appears to leave a lasting imprint.
The benefits also tracked with the intensity of the experience. Participants who showed the greatest increase in flexible, varied brain activity reported more personal insight during the session and, later, greater improvements in their sense of well-being. The correlation supports a leading theory that the psychedelic experience itself — the temporary loosening of the brain's habitual patterns — may be what helps people break free from entrenched, rigid ways of thinking that underlie conditions like depression, anxiety and addiction.
The authors cautioned that the trial was small and conducted in healthy volunteers rather than patients, and that much more work is needed before psilocybin could be prescribed. But by capturing, almost in real time, how a single dose reorganizes brain activity and how long those effects endure, the study helps explain a growing body of clinical results and strengthens the case for the psychedelic as a genuinely new kind of tool in psychiatry.
